Applications

The cyclotron consumable that gates every F-18 batch downstream

[18F]FDG production

The default target water for [18F]FDG synthesis — the F-18 from 98–99 % O-18 water feeds the FDG synthesis module with the activity profile FDG-grade kits expect. Higher enrichment means higher per-shot F-18 activity for the same beam time.

[18F]NaF skeletal imaging

F-18 production for [18F]NaF skeletal-imaging tracer. NaF formulation is a downstream radiochemistry step from the same target water — same isotopic spec, same vial spec.

[18F]PSMA + neuro tracers

F-18 production for emerging clinical tracers — [18F]PSMA-1007, [18F]Flutemetamol, [18F]Florbetapir and [18F]choline. High-enrichment target water gives sharper per-shot activity for the small-volume target-loading workflow these tracers use.

Investigator-initiated F-18 work

IIP F-18 labelling and method-development workflows — same target water, same isotopic spec. The CoA serves as the input-material qualification record for the investigator protocol.

O-18 water recovery + recycling

After F-18 elution, the post-irradiation target water can be ion-exchanged + concentrated for re-use. Higher-enrichment input water means higher-enrichment recovered water — supports closed-loop O-18 water economics for high-volume sites.

Cyclotron target validation

On a new target body or after a target rebuild, the target validation run uses certified-spec water — eliminates the input-material variable when characterising target performance.

Why O-18 Enriched Water?

98–99 %
Atom O-18 enrichment

The 18O(p,n)18F cross section scales with the O-18 atom fraction. Moving from 95–97 % entry-grade water to 98–99 % cyclotron-grade gives a measurable per-shot activity boost — same beam time, more F-18 at end-of-bombardment, more patient doses per batch.

< 3.0 µS/cm
Conductivity gate

Ionic impurities in target water co-irradiate with the O-18 and produce parasitic radionuclides (Na-22 from Na, Co-58 from target-body contact). The < 3.0 µS/cm conductivity spec gates the input material below the threshold where these by-products show up on the QC gamma spectrum.

CoA per lot
Inspection-ready

Per-batch certificate of analysis is the input-material qualification record for the cyclotron GMP file. AERB inspection of a PET radiopharmacy reads the CoA chain; cold-chain delivery preserves the chain from manufacture to target loading.

FAQ

O-18 enriched target water — questions we hear from Indian cyclotron teams

What O-18 water is and why F-18 production depends on it, why 98-99 % atom O-18 rather than lower enrichment, what the CoA and cold-chain deliver for the GMP file, and O-18 water recovery service.

Q.What is O-18 enriched water and why does a PET cyclotron need it?

O-18 enriched water ([18O]H₂O) is water where the oxygen atoms have been chemically enriched from natural-abundance ~0.2 % O-18 to 98–99 % O-18. It is the target material for F-18 production — the cyclotron beam hits the O-18 water target, the (p,n) nuclear reaction converts O-18 to F-18, and the enriched water solution now carrying the F-18 activity leaves the target for the synthesis chemistry. Without high-atom-percent O-18 water, F-18 yield drops sharply and per-patient F-18 cost becomes uneconomic.

Q.Why 98–99 % atom O-18 rather than lower enrichment?

F-18 production yield scales with O-18 atom fraction — using 90 % atom O-18 water instead of 98 % cuts the F-18 activity per target load by roughly 8 %. Over a year of production at a busy PET centre, that difference is thousands of GBq of missed F-18 activity and hundreds of patients that could not be scheduled. Higher atom percent is more expensive per gram but pays back through the higher F-18 yield per beam-hour.

Q.How is the water supplied and quality-checked before target loading?

Each lot ships with a Certificate of Analysis documenting atom percent O-18, chemical purity, sterility and any trace-contaminant analysis. Cold-chain delivery preserves the material from manufacture to target loading. The radiopharmacy verifies the CoA before loading the target — that CoA becomes the input-material qualification record for the cyclotron GMP file that AERB inspection reads.

Q.What is the shelf life and storage requirement?

O-18 enriched water is chemically stable at room temperature; the shelf-life is set by the sterility qualification of the vial rather than by chemical degradation. Typical shelf life is 12–24 months depending on the vial format; the CoA specifies the exact expiry for each lot. Recovered / recycled O-18 water (the un-reacted majority of the target loading) can be re-purified and re-used through the manufacturer's recovery service — significantly reduces per-batch running cost for high-throughput centres.
